Summary: Exposure to only one of these obesogenics, tributyltin (TBT) or the high carbohydrate diet (HCD) alone, can cause obesity and reproductive complications such as the features of premature ovarian failure (IOP). However, little is known about the interactions between TBT and HCD, and the effects of combined exposure on reproduction. In this study, we evaluated whether acute exposure to TBT combined with HCD results in reproductive and metabolic irregularities. Wistar female rats were treated with TBT (100 ng/kg/day) and HCD for 15 days, metabolic and reproductive outcomes were evaluated. The TBT and HCD experimental groups exhibited metabolic impairments, such as increased adiposity, abnormal lipid profile, alteration in the triglyceride and glucose (TYG) index, and adipocyte hypertrophy in HCD-TBT rats. These metabolic consequences have been associated with reproductive disorders. Specifically, the HCD-TBT group exhibited irregularities in the estrous cycle, increased levels of follicle-stimulating hormone (FSH), reduced levels of anti-mullerian hormone (AMH), reduced ovarian reserve and the number of corpora lutea (CL), with an increase in the number of atresic follicles, suggesting that the combined exposure to HCD-TBT exacerbated the characteristics of IOP. Furthermore, strong negative correlations were observed between adipocyte hypertrophy and ovarian reserve, CL number and AMH levels. Combined exposure to HCD-TBT further resulted in reproductive tract inflammation and fibrosis. Collectively, these data suggest that combined TBT exposure to HCD leads to metabolic and reproductive abnormalities, exacerbating the features of IOP in Wistar female rats.
